含二氟甲基的稠杂环化合物合成
摘要
有机分子中的CH键换为CF键会对原来分子的物理、化学和生物学性质产生明显变化,因此含氟有机分子在药物化学、天然产物化学和农药化学等领域有着独特的应用。杂环化合物在现代生活中有着非常广泛的用途,因此把氟元素引入杂环化合物的研究备受重视,而且取得了一些重大进展,一些含氟杂环化合物已经在工业、农业、医药上得到广泛的应用。本课题研究如何高效地将二氟甲基引入到喹啉或者异喹啉上,进而生成含二氟甲基的稠杂环化合物,该方法将进一步丰富有机合成方法学体系,同时会在药物合成中有着潜在的应用价值。本论文中,我们采用无金属参与的反应体系,在可见光促进的有机催化条件下,通过对温度、催化剂、溶剂等反应参数进行考察,实现了稠杂环化合物的二氟甲基化反应。此反应能够以好的收率得到含二氟甲基的稠杂环化合物。基于实验结果和已有的文献报道,我们提出了稠杂环化合物的二氟甲基化反应的可能反应机理:该反应可能经历了自由基加成/分子内串联环化。

in the fields of medicinal chemistry, natural product chemistry, and pesticide chemistry. Heterocyclic compounds have a very wide range of uses in modern life. Therefore, the study of introducing fluorine into heterocyclic compounds has attracted much attention, and some major progress has been made. Some fluorinecontaining heterocyclic compounds have been widely used in industry, agriculture and medicine. This topic studies how to efficiently introduce difluoromethyl groups to quinoline or isoquinoline to generate difluoromethyl groupcontaining fused heterocyclic compounds, which will further enrich the organic synthesis methodology and will have potential in drug synthesis. In this paper, we have developed a reaction system without metal participation and under the visible light redox catalysis, torealize the difluoromethylation of fused heterocyclic compounds. Based on the experimental results and existing literature reports, we proposed a possible reaction mechanism for the difluoromethylation reaction of fused heterocyclic compounds: the reaction may have undergone free radical addition / intramolecular tandem cyclization.

引言 有机分子中的CH键换为CF键会对原来分子的物理、化学和生物学性质产生明显变化,因此含氟有机分子在药物化学、天然产物化学和农药化学等领域有着独特的应用[1]。有机分子中引入氟原子或含氟基团后,会导致分子的整个电子云发生偏移,分子的偶极矩、酸碱性会受到影响,相邻基团的性质和分子构型也会发生变化,进而影响有机分子的理化性质。氟原子或含氟基团的引入,会增加分子的脂溶性,可以提高分子在生物体内的吸收和传递。此外,氟氢键的形成也会对分子的性质产生影响,正是由于以上这些特殊性质,使得含氟化合物表现出优越的理化性质和生理活性。
