Abstract:The rumen of the goat is the main site of material metabolism, with holds a large number of complex microorganisms. With the development of animal husbandry in China and the improvement of people’s living quality, the demand for dairy products has increased greatly. In order to improve the milk performance of dairy cows, it’s a common strategy to feed lactating dairy cows with higher grain diets. However, feeding a high grain diet easily leads to the abnormal fermentation in rumen with high accumulation of free volatile fatty acids (VFA) and then to decrease the ruminal pH value. Subacute rumen acidosis (SARA) was usually induced by feeding a high grain diet. It will eventually reduce the milk performance. In this study, Seventeen midlactating dairy goats were randomly allocated to 3 groups in this study. The control treatment was fed a lowconcentrate diet (35%concentrate, n=5, LC) while there were two high concentrate treatments (65% concentrate, HC), one fed a high concentrate diet for a long period (19 wks, n = 7, HL) and the other fed a high concentrate diet for a short period of time (4 wks, n = 5, HS). Our results showed that ruminal fermentation was changed by HC diet feeding indicating by VFA concentrations and the ratio of acetate/propionate, as well as endotoxin concentration in the ruminal fluid.

随着我国畜牧业的发展和人民生活水平的提高,乳制品的需求量大大增加,生产上为了提高动物的产奶量,通常饲喂含精料较高的日粮,来提高动物的生产性能,但这种方式会使反刍动物瘤胃发酵产生过多的挥发性脂肪酸,导致瘤胃PH下降,诱发亚急性瘤胃酸中毒(Subacute ruminal acidosis,SARA)。瘤胃是反刍动物特有的器官,供给合成乳脂重要的前体物质,罹患SARA时,瘤胃发酵蓄积的大量酸性物质破坏了瘤胃组织上皮细胞,引起炎症,粘膜坏死脱落,进而使瘤胃壁通透性增高,细菌脂多糖容易进入血液,最终引起全身性的炎症反应。
研究表明,日粮精粗比影响瘤胃VFA及乳产量,合理精粗搭配有利于乳脂率的稳定,长期饲喂高精料日粮可在一定时期内可提高乳产量,但会降低泌乳期山羊乳腺组织的抗氧化能力,同时可降低乳中乳脂率以及乳腺组织中乳脂,乳糖合成相关基因的表达水平,使乳的品质下降[1],也有研究认为产气饲喂高精料日粮会激活急性反应和炎症反应从而导致肝脏中营养分配和能量再分配,导致奶制品质量下降[2]。且长期饲喂奶山羊高精料日粮会使LPS和挥发性脂肪酸蓄积在消化道中,诱发亚急性瘤胃酸中毒(SARA),SARA是由喂食过量高度可发酵饲料和不足量粗纤维膳食引起的代谢性疾病[3,4]。临床上通常用几种瘤胃pH值(例如,6.0,5.8,5.6和5.5)来定义SARA[5,6,7],Fairfield等[8]研究表明约20%的商业化奶牛牧场中泌乳早期及中其奶牛瘤胃pH<5.5,出现亚急性瘤胃酸中毒状态。而SARA会导致乳产量和乳脂率下降,脂代谢、蛋白代谢、乳蛋白含量发生变化[9]。国外研究表明,饲喂高精料引发的瘤胃酸中毒与蹄叶炎有关[6],但相关发病机制尚不清楚。目前一般认为高精料经瘤胃发酵产生的大量LPS等有毒物质经血液循环到达蹄部,引起了炎症反应,从而导致蹄部缺氧、缺血、坏死。高精料发酵产生的挥发性脂肪酸(Volatile fatty acids,VFA)及内毒素(LPS)、组胺等有害物质会减弱瘤胃的蠕动,导致采食量下降[8,9]。以发病快、病程短急、全身症状加重、死亡率高为特征。国内研究表明,长期饲喂高精料日粮导致泌乳期山羊NEFA,AST和葡萄糖含量升高,以及肝脏、肌肉和脂肪组织中参与糖异生关键基因的表达上调,提示机体代谢紊乱及糖异生增强[2]。另外,长期饲喂高精料日粮,还会引起羊肠黏膜损伤,主要是通过增加上皮细胞凋亡和炎症反应。[10]研究瘤胃代谢的方法有气相色谱法,体外培养瘤胃液检测发酵产物学等方法。研究瘤胃代谢的方法有荧光定量PCR检测微生物菌群含量,体外培养瘤胃液检测发酵产物,代谢组学测试,分子生物学等方法。本课题通过系统研究饲喂高精料日粮对山羊瘤胃代谢与功能的影响及机制研究,为防治SARA的研究提供更多理论依据。
