Construction and Characterization of ChzMTolA Gene Knockout Mutant in Streptococcus Suis CZ130302 Strain

Abstract:From March to May 2013, pandemic meningitis was spread in Jiangsu province. The strain CZ130302 was identified as the S. Suis. the current research on the strain CZ130302 is less. To research the function of ChzMTolA gene in CZ130302, the isogenic ChzMTolA mutant was constructed. To research the function of ChzMTolA gene, the biological characteristics and pathogenicity of △CZ130302ChzMTolA and wild type CZ130302 were compared. The result showed that their colony morphology, hemolytic and growth characteristics were no obvious difference, the Hep 2 cells adhesion of △CZ130302ChzMTolA dropped 36.69%, the bEnd3. Cells adhesion of △CZ130302ChzMTolA dropped 31.27% ,the toxicity of △CZ130302ChzMTolA declined obviously, the survival rate of BALB/c up to 50%. ChzMTolA gene played an important role in pathogenicity of CZ130302.

猪链球菌(Streptococcus suis, SS)属于革兰氏阳性、兼性厌氧型、具有溶血性、单个或散在的球菌,液体培养基培养时呈短链状,是重要的人畜共患病病原之一。SS是引起世界范围内猪链球菌病的最主要的病原,它不但可以引起猪的败血症,侵害其中枢神经系统和其他组织器官,引发脑膜炎、肺炎、心内膜炎和关节炎[13]等炎性反应,给我国养猪业造成严重的经济损失,还可引发人的化脓性关节炎和中毒性休克综合症[45],对公共卫生和食品安全构成重大威胁,是一种引起公共卫生安全问题的重要病原。
